OECD TG 301D: Closed Bottle Test (Ready Biodegradability)

Understanding the degradation rate of a chemical in aquatic environments is the first step toward regulatory compliance and environmental stewardship. We offer OECD TG 301D compliant testing services under GLP standards, helping you verify the biodegradability of your products and demonstrate a commitment to sustainability.

Study Objective

To determine the biodegradability of a test substance by measuring the consumption of dissolved oxygen (DO) by microorganisms in a closed system over 28 days. The study identifies whether a substance is classified as "Ready Biodegradable."

Service Highlights & Technical Specifications

  • Broad Applicability: Specifically designed for volatile, poorly soluble, or insoluble organic substances by preventing losses to the atmosphere.

  • High-Precision Monitoring: Dissolved oxygen levels are measured at specific intervals (e.g., Days 0, 7, 14, 21, and 28) using high-sensitivity oxymeters to ensure an accurate degradation curve.

  • Strict Quality Control: Inclusion of blank controls, reference substances (e.g., Sodium Benzoate), and toxicity checks to validate inoculum activity and test reliability.

  • Eco-labeling Support: Results serve as critical data for environmental hazard classification (GHS) and green product certifications.

Study Procedure

  1. Inoculum Preparation: Microorganisms are typically derived from activated sludge from a municipal sewage treatment plant or surface water.

  2. System Setup: The test substance and mineral media are placed in completely full, airtight bottles to exclude any headspace or air interference.

  3. Dark Incubation: Samples are incubated in the dark at a constant temperature ($20 \pm 1$°C) for a duration of 28 days.

  4. Data Evaluation: The percentage biodegradation is calculated as the ratio of Biochemical Oxygen Demand (BOD) to Theoretical Oxygen Demand (ThOD). A pass level of 60% ThOD within a 10-day window defines "Ready Biodegradability."
